The following VB.NET code sample loads a large image file (105MB in my test) and saves out an 8KB 50x50 thumbnail.  This took about 8 seconds to run on a n AMD 1500 GHz with 512 RAM.  Of course your 500MB file will take longer, but if it is has multiple pages and you only want to make a thumbnail of the first page, it will not take as long.

        img = codecs.Load("D:\test images\really big.tif", 0, CodecsLoadByteOrder.BgrOrGray, 1, 1)

        'change image size
        img2 = New RasterImage(RasterMemoryFlags.Managed, 50, 50, img.BitsPerPixel, img.Order, img.ViewPerspective, img.Palette, Nothing)
        Dim r As New ResizeCommand(img2, RasterSizeFlags.Normal)

        codecs.Save(img2, "d:\junk folder\thumb.tif", RasterImageFormat.Tif, 24)
